Seeburg R mra5-l6 voltage testing

Can the 5U4 tube be powered on without the mech attached in order to test amp voltages? Receiver and amp have been re-capped. The mech is currently disassembled as part of restoration.

I would NOT power up an "unloaded" rectifier simply because sustained peak DC voltage could potentially stress & damage the electrolytics down the line.
And having "part" of the associated equipment disconnected (mech) can influence voltage readings anyway, giving false readings that are different (higher) from those listed in the service manual.
